Gene expression shapes the safety and efficacy of any cell or gene therapy. Standard regulatory sequences including viral promoters often do not give you the control your program needs. Annogen has a decade of experience, developing and validating expression systems through screenings performed directly in relevant vitro and in vivo models. Our promoters are used by leading biopharma companies, and some are already used in clinical stage therapies.
Primarily promoters, but the same approach can be applied to enhancers, introns, silencers, 5' and 3' UTRs and even viral capsids, and we have experience with each of these. The power of our SuRE platform resides in its demonstrated broad applicability (AAV, Lenti, PiggyBac; human, animal, insect, plant) and high sensitivity, allowing Annogen to distinguish signal from noise to select the elements that truly stand out. We have a strong record of improving the starting element, though the achievable gain depends on the additional demands with regards to cell type specificity or inducibility, for example: We include standard benchmarks such as CMV, CAG or EF1a as internal references in every screen, so performance is always measured against a known baseline.
Yes, and this is a common need where a single promoter must do several jobs. We can screen for elements that are restricted to the right cell type, respond to activation, or stay effectively silent until the target cell state is reached, rather than stacking multiple promoters. Of course, each additional demand can compete with other demands so that for example the lower the expression needs to be in an off-target cell type, the harder it may become to still have exceptionally high expression in the on-target cells.
We have off-the-shelf families including housekeeping, T-cell, liver, cardiac, and neural promoters. For targets not yet in the catalogue, including tissues such as kidney or eye and cell types such as NK cells, we can run a bespoke screen. A typical work package runs about three months, and a full bespoke project including in-vivo work can take up to around nine months. We structure projects in stages, so you see results before committing to the next one. At the end, you receive a set of top candidate elements, typically around five validated sequences, that you can take forward and protect as your own IP.
